One of the many things I like about Charlie Musselwhite's playing is the little effects he throws in as he moved from riff to riff to keep it all n interesting.
On Chicken Shack, off Stand Back, 1966, , openingverse, on the IV chord, there's a little "thunk" before he plays the riff.
Hopefully you hear what I'm referring to.
Does anyone know how to play that?

I think that the "thunk" you're referring to is just Charlie's throat making the noise as he begins his attack of the note and it's being picked up by the mic.

I remember stealing that album from my older brother when I was a kid, 1967 or so. I still have it. (The round vinyl spinnie thing)
Now the album is in my itunes as 1's and 0's. Some cool licks here and there to grab.
Anyways, here's the tune, for reference. And I concur with Kingley. Added: It's what Adam refers to as a "cough"

His harmonica according to CM record/cassette/CD is A+++ and the book is a treasure chest of pertinent info and tab :)

Anyone know where to find that book? I tried once and only found an instructional book that he and Phil Duncan did.
