I've got a 1954 Fender Tweed Deluxe. It currently has a Mojo Tone speaker in it. (I installed it many years ago.) I'm thinking about swapping it out for something more like the original Jensen P12R. What's the difference between the original P12R and, say, one made for a 1957-58 Deluxe? Is there any appreciable difference? If I decided eventually to sell the amp, would a collector care about the year difference between amp and speaker, as long as it was a Jensen P12R?

From my experience owning dozens of 50's Jensens with many still in use. There shouldn't be any difference.
A collector is more attracted to an amp with a date correct speaker even though it may not be original to the amp. The fact that you don't have the original speaker anyway, isn't going to devalue the amp from what it already is. A real P12R will increase the value over a Mojotone. I have a couple 50's P12R's not being used, one is a 55 and one is a 1960, I may part with one if the price is right.

Collectors always like the original, but having a speakers from 57-58 Deluxe is the next best thing.
