How many screws should I use replacing nails? More specifically, should the cover screws (I'd prefer 2) also serve to replace the 2 reedplate nails that align with the coverplate nails?
Thanks!

Do you mean 2 cover screws total or 2 on each end. I replaced the cover plate nails on my Blues Harps with screws, using 2 on each end. Probably over kill, but that's what I did. Just be carefull not to over tighten.

2 on each end. Yes, I do know about 1vs2 on the ends.

Filisko uses three reedplate screws. Stay away from drilling through the really narrow area behind the longest slots. I's far too easy to break the comb in two pieces drilling there.

For the cover plate screws, I use 1 at each end at the front of the harp. I also use 3 reed plate screws at the back for the harmonica. If the comb is sanded nice and flat you do not need a lot screws to get it air tight. The are simply there to hold it together.

There is some good information in this series of videos on the subject. (I have posted the first one below).
